Using PCR, it is possible to replicate a single strand of DNA such as an STSs millions of times. Nucleotides tagged with 32P (radionucleotides) or fluorescent dye can be used along with untagged deoxyribonucleotide phosphates (dNTPs), allowing researchers to denature the STS and make probes for genomic or cDNA libraries.
