Hydrolysis of cisplatin

PtCl2(NH3)  + H2O  =  PtCl(NH3)2H2O+ + Cl-

Binding occurs in cells where chloride concentration is low.

 

The cis isomer, after hydrolysis of the Cl groups, binds to two side-by-side guanines on DNA.

The base guanine to the left and the base on the DNA backbone to the right.
